Such changes in gain possibly result from changes in compliance of intrafusal fibers. Isolated spindles have been used to study these changes. One spindle pole was attached to a tension transducer and stretch applied to the other; receptor potential was recorded from the primary ending. Receptor potential response to small amplitude sinusoidal stretch fell abruptly after a large stretch and recovered over a period of several sec. Following direct stimulation of the intrafusal bundle through Ag: AgCl wire electrodes, a residual increase in tension was observed. This was abolished by a large stretch; it appears related to the after-effects of fusimotor stimulation. In other experiments decapsulated isolated spindles were observed with Nomarski optics. Depolarizing current in the bag1 fiber caused contraction followed by a smaller residual sarcomere shortening; this may also be related to the after-effects of fusimotor activity. Measurement of length changes with stretch shows that, for small stretches, intrafusal fibers are relatively more compliant in the sensory region than elsewhere. The SHV family of b-lactamases appear to have been derived from Klebsiella spp. The progenitor of the SHV class of enzymes, SHV-1, is universally found in K. pneumoniae. In many strains of K. pneumoniae, the gene encoding SHV-1, or its apparent precursor, LEN-1, resides within the bacterial chromosome too; it may be that the gene for SHV-1 b-lactamase evolved as a chromosomal gene in Klebsiella and was later incorporated into a plasmid which has spread to other enterobacterial species. SHV-1 confers resistance to broad-spectrum penicillins such as ampicillin, ticarcillin and piperacillin but not to the oxyimino substituted cephalosporins.2 Then, in 1983, three strains of K. pneumoniae and one Serratia marcescens isolated in West Germany were demonstrated to cause transferable resistance to cefotaxime, as well as to other newer cephalosporins.5 This new plasmidic b-lactamase, called SHV-2, derived from a mutation in SHV-1. Florida's current attempt to control prescription drug spending is unlikely to save money and is a dangerous step toward putting government, rather than doctors and patients, in charge of the patient's health care priorities. Florida lawmakers may be tempted to respond to their failure to control costs by enacting further restrictions on access to medications. SBSE ; technique. We optimized and compared them in order to establish which is better for determination of volatile compounds in grapes. We determined several major aroma compounds: benzaldehyde, linalool, -terpineol, nerol, geraniol, hexanal, nonanal and decanal. The extracted compounds were identified by mass spectrometry GC-MS ; . Standard solutions were prepared in methanol and n-hexane. Solid phase microextraction was performed from headspace HS-SPME ; and five SPME fibers were tested. DVB CAR PDMS divinylbenzen, carboxen, polydimethylsiloxane ; fibre was found to have the best characteristics and was used to optimise the extraction temperature and time. In Fig. 1. we show an example of optimisation of extraction time at 50 C. For all compounds except hexanal the best results were obtained at 15-min thermostating at 50 C and 35-min extraction time. The optimal extraction conditions for hexanal were at room temperature for 15 min. To optimise the analytical procedure, we used commercially available white and red table grapes of a non-specified variety. After maceration and under the same conditions, more aroma compounds were found in the pulp of red grapes with the approximate ratio 1.5: 1 for red and white grapes, respectively. In both grapes the main compound was hexanal with the concentration cca. 5 mg L. The skins were separated from the pulp for both types of grape and analyzed separately. A higher concentration of aroma compounds was determined in the skins. We found more hexanal in the skins than in the pulp: 56% more in white grapes, and 64% more in skins of red grapes. The concentrations of other compounds were cca. 11% higher in skins than in the pulp for both grapes. The stir bar sorptive extraction SBSE ; technique was also tested. The advantage of this technique over SPME is a larger surface area of the exctraction phase and thus a higher capacity. In this case the extraction phase was polydimethylsiloxane PDMS ; therefore the extraction efficiencies depended on the polarity of volatile compounds. The extraction was carried out at room temperature using a magnetic stirrer, while the desorption was made into n-hexane using an ultrasound bath and not in a SBSE desorption unit. The conditions for this technique were also optimized.
